For many workflows, especially those requiring isogenic and gene edited cell lines, it is essential to generate a truly clonal cell line – that is, derived from a single cell.
For workflows requiring high numbers of clones, for example, multi-gene editing, this requirement for a clonal iPSC cell line represents a significant challenge.
iPSCs are sensitive and labour-intensive to culture. Traditional methods such as limiting dilution or colony picking yield low numbers of clones for downstream evaluation. With little or no documentation on the clonal origin of a cell line, this can become a real issue for researchers needing to satisfy regulatory requirements in the future.
